HPLC Peak Tailing: 7 Causes, Fixes, and How to Choose a Column That Prevents It

You inject a standard you have run a hundred times, and the peak that used to be a neat Gaussian now drags a long tail behind it. Integration becomes unreliable, resolution drops, and a method that passed system suitability last month suddenly fails. Peak tailing is one of the most common HPLC problems, and one of the most fixable once you know where to look.

What counts as peak tailing?

Tailing is measured with the tailing factor (Tf), calculated as the peak width at 5% height divided by twice the distance from the peak front to the peak maximum at that height. A perfectly symmetrical peak gives 1.0. Many methods, including those following the USP <621> system suitability, allow values up to about 2.0, but plenty of labs set a tighter internal limit around 1.5. Check what your own method requires.

7 causes of HPLC peak tailing and how to fix them

1. Silanol interactions. Residual silanol groups on the silica surface can hold onto basic analytes a little longer than the main reversed-phase mechanism does, and that delay shows up as a tail. Lower the mobile phase pH to around 2.5-3 so the silanols are mostly protonated, add a proper buffer, or move to a high-purity, fully end-capped column.

2. Mobile phase pH too close to the analyte pKa. When pH sits near the pKa, the compound exists in two forms at once, and the peak smears. Stay at least 1.5-2 pH units away from the pKa wherever your column's pH range allows. Our guide to mobile phase preparation covers how to set and verify pH correctly.

3. Weak buffer. A buffer at 2-5 mM often cannot hold the pH at the point of injection. Try 10-25 mM for UV methods. For LC-MS, keep it volatile and lower.

4. Column overload. If the peak gets more symmetrical when you dilute the sample tenfold, you were overloading the column. Reduce the injection volume or concentration.

5. A void or blocked inlet frit. Particulates from samples or pump seals build up on the inlet frit, and a settled bed leaves a gap at the head of the column. Both distort every peak. If the manufacturer allows it, back-flush with the detector disconnected. If that does not help, the column has reached the end of its life. A guard column is the cheapest way to stop this happening again.

6. Extra-column volume. Wide tubing, long connections, or a fitting that is not fully seated create dead volume that tails early-eluting peaks most of all. Use narrow-bore tubing appropriate to your system and re-make suspect connections. Our comparison of PEEK vs stainless steel fittings and tubing will help you pick the right parts.

7. Sample solvent stronger than the mobile phase. Dissolving a sample in 100% acetonitrile and injecting it into a 20% acetonitrile mobile phase makes the analyte race ahead of the band. Match the sample solvent to the starting mobile phase, or inject a smaller volume.

Choosing a column that resists tailing

If you analyse basic compounds regularly, the column matters more than any additive. Look for high-purity (Type B) silica, full end-capping, and a pH range that suits your method. Hybrid particles tolerate higher pH, which can neutralise basic analytes altogether. When to replace the column instead

If tailing appears together with rising backpressure, shifting retention, or split peaks, cleaning will only delay the inevitable. Frequently asked questions about HPLC peak tailing

What is an acceptable tailing factor in HPLC?

A tailing factor of 1.0 is perfectly symmetrical. Most methods accept values up to about 1.5, and many pharmacopoeial system suitability tests allow up to 2.0. Always use the limit written in your own validated method, since it overrides general rules of thumb.

Why do only basic compounds tail on my C18 column?

Basic analytes, especially amines, carry a positive charge at low and neutral pH and interact with residual silanol groups on the silica. This secondary interaction releases molecules more slowly than the main reversed-phase mechanism, which creates the tail. A lower pH, a stronger buffer, or an end-capped column reduces it.

Can a guard column cause peak tailing?

Yes. A contaminated or poorly connected guard column can add dead volume or trap material that distorts peaks. Replace the guard cartridge first when tailing appears suddenly, and make sure the connections are fully seated.

Does peak tailing mean my column is dead?

Not necessarily. Tailing can come from the mobile phase, sample solvent, overload, or system plumbing. If a known-good column gives symmetrical peaks and yours does not, and cleaning does not help, then the column has likely reached the end of its life.

Is peak tailing the same as peak fronting?

No. Tailing means the back half of the peak is stretched, while fronting means the front half is. Fronting is most often caused by sample overload or a sample solvent that is much stronger than the mobile phase, so it needs a different fix.

Which HPLC column is best for reducing tailing with basic compounds?

Choose a column made from high-purity silica with full end-capping, or a hybrid particle that tolerates a wider pH range. Share your method and analytes with our team, and we will recommend options from Merck, Supelco, ACE, Waters, Agilent, or Thermo.
