LiChrosorb RP-18 HPLC Column: Complete Guide to Features, Uses, and Selection

Reversed-phase HPLC is the workhorse of modern analytical labs, and the C18 column is its most familiar tool. Among the many C18 options available, LiChrosorb RP-18 has earned a lasting place. Many validated pharmaceutical, food and environmental methods were built on it, and labs still search for it by name when they need reproducible results.

This guide explains what LiChrosorb RP-18 is, where it performs best, how it compares with related phases, and how to keep it working well. Whether you are transferring an existing method or choosing a column for a new one, you will find practical points to help you decide.

What Is LiChrosorb RP-18?

LiChrosorb RP-18 is a reversed-phase packing made by Merck. It consists of irregularly shaped silica particles chemically bonded with octadecyl (C18) chains. The silica base has a nominal pore size of 100 Å, and the packing has been offered in several particle sizes, most commonly 5, 7 and 10 µm.

In reversed-phase chromatography, analytes are separated by their interaction with a hydrophobic stationary phase using a polar mobile phase, typically water mixed with acetonitrile or methanol. The long C18 chain provides a strongly hydrophobic surface, so compounds with greater non-polar character are retained longer. Specifications such as surface area and carbon load can vary by grade, so always confirm the details on the datasheet for your exact part number.

Key Features of LiChrosorb RP-18

Strong, predictable retention. The octadecyl phase offers high retention for non-polar and moderately polar compounds. This gives analysts more room to adjust selectivity by changing the organic solvent ratio.

A long track record. LiChrosorb phases have been cited in methods for decades. If your method names this column, using it avoids the time and cost of revalidation.

Robust irregular silica. Irregular particles are cost-effective and mechanically sturdy, which suits routine QC work as well as semi-preparative applications where loading capacity matters.

Wide solvent compatibility. Like other silica-based C18 packings, it works with common aqueous-organic mobile phases within the manufacturer’s recommended pH and temperature limits.

Typical Applications

LiChrosorb RP-18 is a general-purpose column, which is why it appears across so many industries:

  • Pharmaceutical analysis: assay of active ingredients, related substances and impurity profiling
  • Food and beverage testing: vitamins, preservatives, sweeteners and colorants
  • Environmental analysis: residues and organic contaminants in water and soil extracts
  • Method transfer: reproducing legacy or compendial methods that specify this packing
  • Teaching and method development: a dependable baseline column for learning reversed-phase principles.

Choosing between related phases is easier when you know what each one is designed to do.

RP-18 or RP-8? The difference is chain length. C8 is less hydrophobic, so analytes elute sooner and need less organic solvent. If your compounds are too strongly retained on C18, or you want shorter analysis times, RP-8 is a sensible alternative. If you need maximum retention and resolution, stay with RP-18.

What about RP-select B? This phase was developed for challenging separations, particularly basic compounds that can tail on conventional silica. If peak shape is your main concern, it is worth evaluating alongside RP-18.

How to Choose the Right Column

  1. Start with your method. If a validated procedure specifies LiChrosorb RP-18, order that exact packing, particle size, and dimensions.
  2. Check particle size and pressure. Smaller particles give higher efficiency but higher backpressure. Make sure your system can handle it.
  3. Match the column to the analyte. Highly non-polar compounds may suit C8, while mixtures with a wide polarity range often work well on C18.
  4. Think about the sample matrix. Dirty samples shorten column life. A guard column is a low-cost protection. Our article on guard columns explains when you need one.
  5. Confirm dimensions. Length and internal diameter affect resolution, run time, and solvent use.

Troubleshooting Common Problems

Even a good column can show problems if the system around it is not in order.

Shifting retention times. Changes in mobile phase composition, temperature, flow rate, or column equilibration are common causes. Our guide to retention time drift in HPLC walks through eight causes and fixes.

Unexpected peaks. If extra peaks appear in blank runs, the column may not be at fault. Read about HPLC ghost peaks to trace contamination from solvents, vials, or carryover.

High backpressure or broad peaks. A blocked frit, particulate buildup, or poorly made connections can all contribute. Review your HPLC fittings and tubing to make sure dead volume and leaks are not hurting performance.

Tips to Extend Column Life

  • Filter every sample and mobile phase. See our syringe filter guide for choosing pore size and membrane.
  • Stay within the recommended pH and temperature range.
  • Flush buffers out with water and organic solvent before storage.
  • Store the column in the solvent recommended by the manufacturer, with end caps tightly fitted.
  • Avoid sudden changes in pressure or solvent composition.
  • Prevent contamination at the source by following our tips on autosampler vial contamination.

Supporting Consumables

A column performs only as well as the consumables around it. Ekelabshop also supplies vials and caps and a wide range of supplies for chromatography, so you can source your full HPLC setup in one place.
